Synthesis and Capillary Electrophoretic Separation of Polyamidoamine Dendrimers with Ethylenediamine as Core

(CE) 1.0 GPAMAM was isolated and purified to remove 0.5G PAMAM which may be contained therein. 1) In the left beaker, the absorbance of the sample solution gradually decreased, the absorption peak was located at approximately 222 nm, and the peak shape was broad. In the right beaker, the absorption intensity of the sample solution gradually increased, the absorption peak position was approximately 210 nm, and the peak shape was narrow. This shows that capillary electrophoresis has successfully separated the 1.0 GPAMAM from the left beaker into the right beaker.

Mass spectrometry (MS) analysis of the product in the two left and right beakers showed that the left beaker contained 0.5 and 1.0 G of PAMAM, while the right beaker contained only 1.0 G of PAMAM. To obtain pure 1.0 G of PAMAM, first The pH of the system is adjusted so that the ammonium ion (-NH3+) is converted into an amine group (-NH2), which can be separated by HPLC after adjusting the pH to the equilibrium point.

For PAMAM with more degenerate algebra, this method can also be used for separation and purification. However, in the synthesis of high-algebra PAMAM, due to the inevitable existence of some structural defects in the product, it will lead to an increase in the difficulty of separation, and the identity of the final product will be affected.

3 Conclusions The 0.5G-6.0GPAMAM was synthesized by the scattering method at -30 and characterized by nuclear magnetic resonance and elemental analysis. The structural integrity of the product is better than that of the products obtained from similar tests.

Capillary electrophoresis is an effective method to separate and purify PAMAM. This method is not only suitable for the separation of low-algebra PAMAM, but also has reference value for the separation of higher-algebra PAMAM.
